The ideal way to use PoH spam is to go party-by-party. ie PoH Party 1, then PoH party 2, then party 3 etc. In that case you won't be having much of a problem with wasting the glyph procs.
And yes the glyph does overwrite itself.
Now that renew is more usable, haste is stronger. Get to around 12.5-15% then go mastery. That's what I'd do. Remember what your required ratings are depending on Darkness and the 5% raid buff. They're in the stats section of my holy guide at the top of the page. Ignore this advice if you're disc, but your questions sound like holy priest questions.

Learning positioning of your raid members when usin' your PoH helps to make sure you always get your five targets. I usually set one member per party as my PoH 'focus' for each party, and let them know to try and stay centralized with the raid. Helps for ranged groups so you don't end up loosing 1-2 targets due to range issues.
Also the glyph stacks per cast, but expires so fast it won't stack like Holy's Mastery, but it's good to know you don't 'lose' any healing by spamming.

1. It heals the 5 people in the party of the person you're targeting, not your party specifically.
2. No. Serendipity is good for "oh shi-" situations. The two spells that trigger it (Flash Heal and Binding Heal) are both very expensive, and so you won't be getting Serendipity procs very often. It's still a useful talent for when the you-know-what hits the fan, but keeping it up all the time is a recipe for disaster.
3. You just spam it when necessary. I believe the glyph doesn't overwrite itself, although even if it does, it's still worth spamming in most situations. The glyph is just a nice bonus, the real meat and potatoes is the heal itself.
4. Depends on your stats. Mastery is very good, Haste is very good, ideally you'll have copious amounts of both. If you're a fresh 85 working through heroics, I'd suggest Mastery over Haste simply because Haste doesn't help with mana consumption. Once you get int mostly blues, you can seek out more Haste.
There's a whole debate over which of the two is more important for raiding, but everyone agrees that they're both very good. Aim for a good chunk of each.
Wasn't there a time a long while ago when POH could only target your own party? I'm trying to jog my memory but it was during the naxx of wrath that POH was allowed to target other parties, right?
Also, on another note. I would recomend using Binding Heal to stack serendipity since it has more HPM then FH assuming you would also get the full amount of the healing as well. I would agree that maintaining Serendipity all the time isn't worth the effort unless you're popping SoL's like gang busters. I swear, SoL is the Hot Streak of healing.
