Ok cool, but if you couple the Use: bonus with say "standard" buffs from a 10 man raid, and used it on a boss encounter along with any other CD (such as rapidfire, or hero, or any +haste buff) would it produce more dps overall by the end of say... a 5 minute encounter (where it would get used twice)

More - maybe, more relatively to Tia's - no way in hell.
Notice Tia's 340 static AGI is affected by all your cds and raid buffs as well. And they double dips in there.
If an effect can increase usefullness of 1605 agi on 20/120 so it matches 340 AGI it must be an effect increasing the AGI usefullness by 27% (1605*1.

No buff present 100 seconds of 120. Epic trinket gives zero AGI.
Buff present 20 seconds of 120. Epic trinket gives 1605*1.27 AGI.
All 120 seconds in a 120s interval fight. Epic trinket gives 0*100/120 + 1605*1.27*20/120 AGI on average assuming whole fight consist of a parts equal to multipliers of 120.
Its all fine but the effect affects your blue trinket static 340 AGI too, lets say it lasts only 20s and its the exact 20s the epic trinket is activated and it has exact same interval. Now our static 340 AGI all of a sudden is worth 412 AGI**.

All 120 seconds in a 120s interval fight. Tias grace gives 340*100/120+340*1.27*20/120 AGI on average, which equals 355.
So in fact the effect must be 4.5% bigger than previously assumed (355=1.045*340), but 4.5% bigger effect will again affect the static AGI resulting in new net worth of Tia's grace of 340*100/120+340*1.27*1.045*20/120=358, which is slightly (~1%) higher than 355. So again the
effect
must be in fact 1% higher than previously assumed 1.27*1.045. So new effect equals 33% damage increase.
If you find such damage increasing effect that again lasts only 20 seconds out of 120 and you are able to magically fit the trinket in it than yes. Tias grace is equal value to the epic trinket. If the effect is longer or/and Riddle use cant be fit into it each trinket cd, it will need to be more powerful to equalise the fact it still affects Tia's AGI bonus while Riddle AGI bonus is not present.
TL;DR: Tias grace is better.

is macroed (sp) onto
so that it engages pretty much instantaneously every 2 minutes with the added bonus of raidbuffs, LnL and various other limited buffs (wether they proc or not) the overall DPS output is MASSIVELY improved over using
Tia's Grace
. Thats not to say that either item is better, but i would suggest using
Tia's Grace
for 5 mans or mob heavy area's and
Unsolvable Riddle
for raids throughout.

Thats the exact opposite way of using an On Use trinket. The benefit of On Use is being able to stack it with raid buffs as you said earlier like

Heroism
to multiply the effect. Using it on cooldown as you say with no regard for burn phases or cooldown usage is netting you less Agility than using
Tia's Grace
full time. No benefit whatsoever.

I think we are also forgetting to consider one key thing - how much harder it is to get one vs the other. If the damage difference isnt that great, then the easier accessible trinket is better.
Doing exalted with TB is about week, maybe a bit more.
i am running CIty of tolvir on 3 chars each day for over a month, havent seen Tia's grace drop on any of them. Sure i have seen it drop on my mage, bummer.

After even more testing this evening, when i use
Unsolvable Riddle
in this manner i do a burst of an additional 3-5k dps for 20 seconds or so, meaning the overall
damage dealt
over *any* period of time, especially if over 2 minutes is higher than using
Tia's Grace
Best trinket combination regardless is
Prestor's Talisman of Machination
+
Essence of the Cyclone
