is anything you have under your cursor at which you can use/cast a spell on it. It could be from a unit frame (party, raid, battleground), it could be a NPC on which you have your cursor over, it could also be a mob around you during a fight on which you also have your cursor over it.

Putting macros on bars isn't a Bartender specific thing, it's default.
Well done for making it work though, moving from clicking to keybinding can be a tricky process but it will really help I'm sure.

Personally I use something like:
#showtooltip
/cast <ability name>
And just press the keyboard button when my mouse is over the enemy I want to use the ability on.
