As far as I know, you might have to click the macro more than once to activated multiple abilities. I don't have much experience with macro's though, so take my advice with a grain of salt ;]

It's not that it won't hamstring, actually.
It does _attempt_ to hamstring, but when you click a macro it executes it all at once - it won't wait until you are in range from intercept to hamstring.
What you can do is just push the macro once to intercept, then once more when you are in range to hamstring. This also works if you just want to hamstring and are in melee range already - it will attempt to intercept, but it won't be able to because of the minimum range, so it will just use hamstring.

I am pretty sure that an ability used after charge or intercept will work as if it was clicked after you reach your target. You can test by making a charge and demoralizing shout macro. I use that same intercept and hamstring macro and a charge hamstring, zerker stance and both work just fine for me provided I have enough rage for both abilities.

No such thing, the way macros handle the Global Cooldown was implemented to prevent most abilities from being chained together in quick succesion. Some abilities are exempt, though. Notably Heroic Strike, Cleave, Stances, and Bloodrage.

The bracketed part is a condition that tells the script parser when to perform an action. In short, it lets you limit when you want to use an ability. To use your example, it would attempt to use Berserker Rage, but only while not in Defensive stance.
/cast Overpower
/cast Whirlwind
Would attempt to cast Overpower or Whirlwind according to which stance you are in, if you took out the portion you would press the button in Berserker Stance and it would tell you that it cannot cast that spell in Berserker Stance, because it would attempt to cast Overpower, fail, and then stop completely because it has failed. Of course it would be useless to macro this unless you have your bars set up to not swap when you switch stances.
Another common one, allows you to set a target for the ability even if it is not your current target and does not switch your target when doing so.
/cast Intervene
Would cast intervene on your 2's partner,
Someguywhosgettingbeatentodeath
without causing you to lose your current target or forcing you to target him in order to cast intervene.
