From what i understand, Mastery is something only pertaining to the fact that you've spent talent points in large of 1 tree. I.E. Afflic locks will get spellpower as most of their things are insta-cast, and Destro locks will get haste as their spells are mostly casted with time. Whether or not Mastery will be available through gear is still unknown, though from what I've read both here at wowhead and abroad it seems mastery is a talent-only sort of thing. more than likely they will buff the stats of teh actually item itself.

They'll probably turn it into mastery for simplicities sake. And is Expertise really being removed? I can't remember.

I think for the most part, with stats on gear:
AP/SP will be rolled into Agi/Int, possibly buffing just the one or maybe other stats.
MP/5 will be converted to Spirit.
ArP, defense, and block value will be converted to mastery, or possibly other stats (but mastery most likely). If both are on a piece, you will probably see the amount of other stats change.
Haste is listed as a change, not a removal.
So on current gear, the stats will get shuffled around. On new gear, they are obviously creating said gear with the new stats in mind.

I think Mastery points on gear will give stats according to your spec? That's the vibe I seem to be getting from the changes overall, and it sounds great because it will help a #$%^ton with itemization.

So wait, you think Mastery is going to have ArP Def and Block all together? Man... that would be OP for tanks lol.
I meant the stat on gear (e.g. if the gear currently has ArP) would likely be converted to Mastery. The stat itself is going to provide a benefit based on your spec - for example, a Holy Paladin would get extra critical bonus on crit heals (e.g. instead of a crit being 150% heal it may be 164% with 14% mastery), an assassination rogue would get more poison damage, etc.

Don't forget that professions will be able to re-itemize! I could easily see Blizzard doing something like wiping the ArP/MP5/BV/DefR/etc. straight off a piece, not placing the points into anything else, and having X amount of itemization points left over for you to bring to a Blacksmith/Tailor/LW/Engineer/JC to get brought up to current stats.
That won't happen, Blizzard will not require you to get the help of a profession to get the item brought up to the stat value it deserves by its item level. Enchants are in addition to the stat value (e.g. they don't take 55 stats off of a 2H because you can add 110 AP to it with an enchant) and reforging will remove part of a stat to add the other. So if you get a piece that has 100 agi, 150 stam, 100 crit, 100 hit; and you are over hit capped you could convert 50 hit into 50 mastery. But you wont be just arbitrarily adding 50 of a stat.

Mastery gives a spec specific bonus.
Protection Paladins would increase in block percentage (blocks will now prevent a set percentage of damage instead of a flat amount).
That same stat would reduce spell cooldowns on a Retribution Paladin, giving no benefit to blocking (although why a Retribution Paladin is running around with a shield is anyone's guess).

's information regarding mastery. From what we can tell, each talent point you put into a tree results in gaining percentage in 3 stats which are specialized to your spec. One of those three stats will be further enhanced by your mastery stat, which gives you and increase in one of those three stats.
So in the example of a ret pally, you get melee damage, melee crit, and ability cooldown reductions. These stats are not set in stone (the numbers or the whole stat could be changed), and it is entirely likely that it only applies to retribution abilities. If you were prot, your mastery would likely increase block value instead, and it is likely you will see a further reduction in ability cooldown in Prot talents than mastery from Ret, and more overall damage reduction from the prot talents as well.
