Yes, the glyph is definitely worth it. I only hit plague strike on single target fights now (and use Pest to keep both diseases up) to give added threat when hitting with Oblit and Blood Strike. With rime procs and the cooldown on HB, you can spend your blood runes on BB for AoE threat instead of spreading diseases.
The glyph makes AoE tanking far easier now than it is without it. It also means that if you take chailblains as part of your build (like I do), it gives you that extra bit of control when someone pulls aggro off the unmarked target (so many nub dps out there don't understand the concept of 'kill skull').
My three glyphs are HB, FS, and Disease. Frost Strike for 32RP is just too good to pass up. It's the best way to get snap aggro back on a boss that has an aggro reset - keep your RP topped up - when the reset hits, smack away with a few frost strikes. Insta-aggro again. =)

Glyph of Pestilence is a dps loss for all variants of Frost and Unholy at least. If you use HB glyph you use a single disease rotation attempting to refresh FF as often as possible with a HB on Rime procs.

Wait...what? PS only on single target fights? What do you do one multi-target? HB, DnD, ?
Depending on the pull, HB-BB-BB is usually enough snap aggro to keep everything stuck to me. From that point on, OB-FS on the main target, and repeat the above three whenever they're available.
I like to keep DnD ready in case we get adds into a fight. Drop it on the path between the mobs and the healers and they'll turn right back to you (where you hit them in the face with HB and BB again).
The only people who really give me troubles with threat are ranged dps who are fully Naxx10/25 geared and get to the fight before I do.

The only avoidance/tank directed sigil besides the IT one is the
sigil of deflection
from ulduar ,
If you dont have any access to it there's always
Sigil of awareness
or sigil of the
vengeful heart
for tps purposes.

Well if you've got enough Emblem of Valor, Sigil of awareness is your best bet, if you dont, well then the Dk starting sigil is your best bet (since you wont be using DC/SS/IT, making the venture Co. sigils and Unfaltering knight useless, and the Sigil of haunted dreams is just awful :))
