I'm assuming you want two different classes? Nothing will beat two identical classes for ease, and if they're hybrids you can roll different specs at 80. If you're happy with that scenario, either two druids, two shamans or two paladins will work well.
If you're dead-set on two completely different classes, I've heard the melee+caster combination can be difficult to master, mainly due to them needing to be at different ranges (case in point: A priest and a feral druid, priest pulls aggro or gets attacked by a passing mob, cat needs to turn around?)

I dual boxed a priest and mage to 60, double casters make it pretty easy (binded a follow/assist macro key, fireball/mind flay, dot macro/frost bolt, and mind blast/arcane missiles)
I tried for awhile dual boxing a mage and a pally, but the only way I could make it someone viable was to get the paladin to do nothing but heal, but then it slowed down the process (half the possible DPS).
& yes, theres programs that make keys affect both copies of WoW. I personally used Keyclone, but I know theres a whole ton of them that work.

Personally i like the use of a Mage and a Lock. two caster dps in the beginning have similar spells and mana restrictions, Shadow bolt, Ice Bolt a mean combo, not only that but one had ice armor and the other has demonic armor, so you bind all those together and your pretty set, and i make it so i can cast AI on my lock and mage :) Other classes i have tried to DB are: Rogues(Not terribly hard) and Warriors(a little trouble altogether) so i definatly like my Mage/Lock combo
And yes Keyclone is out there it is by far the best/easiest to setup, bad part is it costs 20 bucks, however its a one time purchase and its good forever, until you have to change pc's thentheres a cost to have it transfered. But i love Keyclone, and the best part is unlike stting up macros (Which blizzard is not okay with) Keyclone is legal per blizz's ToS, theyve actually taken the time to speak to Gm's and have SS of the convos. Also all the bindings are already set up you just select which keys you dont want to pass ie: enter for chat etc etc.

I've tried several different setups with my two accounts and Keyclone.
Controlling each char individually
Tried a warrior/priest setup that way, warrior as my "main" and using 1-8 for his actions, priest as "slave" using F1-F8 for his actions. The main trouble with that was keeping them both efficient, I could play the warrior efficiently, but then I would lose track of the cast bar of my priest and he would end up mostly just throwing a heal when needed and being idle the rest of the time. I tried letting my warrior just pick up the mob and after taking aggro just use autoattack while I focused on casting with my priest, but that reduced my warrior to being rather inefficient.
If you go for controlling your chars individually, you'll have to be a master of multitasking or you'll end up with one char "slacking" while the other does most of the work.
Controlling both chars at the same time
Having the main and slave action bound to the same key is much easier. You hit 2 and both your main and slave starts casting. If you pick two different classes, for example warlock-mage, you'll have to pay attention to any difference in cast times. If your main is the warlock and has a 2 sec cast shadowbolt, while your slave mage has a 2.5 sec fireball, the mage won't be ready to cast when you press 2 on the warlock to fire off the second shadowbolt. Ofcourse you can just spam the button like mad and fix it that way, but personally I prefer to make a setup where both chars have similar cast times as much as possible.
Same goes if one class has a cooldown and the other doesn't, for example a druids moonfire bound to the same key as a shammys earthshock, you can spam moonfire, but the shammy will do nothing for the next 5-6 seconds while earthshock is on cooldown or you can limit the druid to only 1 moonfire whenever the shammys earth shock is ready.
The most efficient
..in my opinion is to have two identical casters which spells bound to the same keys. Spells have the same cast time, cooldowns are the same and so on and being casters you don't need to worry too much about getting them into position.
Two different classes CAN work. but you'll risk ending up with one doing most/all of the work and the other acting mainly in a supporting role, meaning that you are pretty much only getting 50% of the potential out of the group.

So I'm lookng at RaFing soon and dual boxing my way to 2 new 80's. I was wondering if anyone out there knows pairs of classes that perform exceptionally well together in pve? I was thinking about Resto Shammy/ Feral druid or Resto shammy/ Lock, but I'm open to suggestions if anyone wants to throw a few out there. :)
Having done this many times, it was how I spun up a full horde setup on a new realm.
Main Account : The class you want to play.
RAF Account : Priest
90% of the time all you need mapped from priest is Shield, Renew, Disease, and maybe Flash Heal.
My setups were always /focus set the same key on both characters, hence I press F2 and the focus key. All priest spells were either target=focus or target=self (or is that player)
Why priest? Shield and Renew are all most classes need in support. Neither is interrupted by movement. While druids appear similar they only have LB (late) and Rejuvenation that can fly with movement.
I always used the RAF for a healer. I made 6 priest that way and had six 61s and 3 60s on the main afterward. (one of the free three 60s was a priest)
Average time to 60/61 is less than 24 hours played. (no instance running either)

If your priest is slacking, set up an assist macro and just wand-spam for the other times.
We're currently experimenting with dual hunters - it's much easier than two different classes.
The other option is to pick classes that don't have much involvement. Paladins make good choices.

We're using HotKey - it's alright? I haven't tried Keyclone, so I don't know how it compares.
Heirlooms make RaF'ing a bit faster, I think, but it's not going to make a huge difference. Quest gear is more than enough.
Just remember to have some cash set aside for your new characters. Nothing burns up cash like having to train, without grinding out quest rewards and farming low level mats.

If you're going to have a "tank" class, I suggest Paladin. Even if it's Ret, you can still turn on Righteous Fury and have significantly high threat. Druid and Warrior's tank stances both have a loss of DPS and a loss of certain DPS abilities. Paladins can still use all their normal abilities, just with higher threat.
If you're going to have an actually heal-specced healer sidekick, either Disc. priests or Shamans have the best "set and forget" abilities.
After next week, finals week, I'll be starting RAF with a Pally and either a Priest or Shaman.
