I'm relatively new to making macros, but I've been reading a little about the mailbox API suff. Here's what I'd love to do: As a guildleader, I use the mail system a lot to send welcome messages/items to new members, and distribute gold/door prizes to members for event participation. I'd love to automate this a little bit. Like for instance, I'd love to be able to have a macro that fills in a predefined subject and message, and attaches a predefined amount of gold and/or item. Is this even possible?

Not with a macro. The character limit of 255 would be way too small. There are mailbox AddOns, such as Postal, but I do not think it can have predefined attachments.
That sounds like a custom AddOn that would only be useful for the purpose you describe. Further, to send normal mail, said custom AddOn would interfere, and you would have to disable it to send normal mail.
It is actually far less work for you to send mail the way you have been doing than to write and test and bug fix a custom AddOn.
Assuming said customization can even be done in the first place...
Well, I found an AddOn that
do the trick, but I haven't tested it, and can't say for 100% certain.
I just want to add in: you don't want to use a macro. Because of the potential for spam abuse, the macro system almost certainly detects thrashing and spamming attempts and will probably disconnect you when sending mails too quickly. You wouldn't be able to elegantly fit throttling logic into the space of a macro. You're much better off using, or designing, an addon around this purpose.
