I am using both. I mean, normal 3 buttons mouse + maybe shift, not enough buttons.
And its not that you have more stuff on your shortcut panel you have to use. I like to utilise all fast react keys.

On a 3 button mouse, using Clique, you can bind an easy 12 skills (Shift, Ctrl, Alt). I believe (but haven't tried it so I could be wrong) that you could use other keys in addition to the mouse buttons as well (z+left click, for example).
You can, but it sure as hell isnt very convenient. And movement with your character is impaired as well.
I use 5-button mouse and still prefer mouseover macros plus clicks combo.
None of the current clickers supports Z + mouseclicks. Only function keys. I dont think it would work anyway - Z + mouse click would mean Z + 1 or Z + something else. You cant press those two buttons together. You could if you remapped alt or control for Z, but thats about it.

I haven't used Clique in a long time (currently using Vuhdo), but can't you also do combos - like Shift+Alt+Button 1? I know you can in Vuhdo, and I thought you could in Clique too. Makes a 3 button mouse actually have 24 skills able to be bound to it.
Then, don't forget wheel up and wheel down - plus all the modifiers, which gives you an additional 16 bindings.
Grand total from a 3 button mouse with scroll wheel = 40 skills bound. That should be plenty.
If we are talking about combinations its probably more.
But it beats the point of having the keybinds at all, because you wont be able to execute them with such ease and such accuracy, while still being able to move and position your character properly as if you had them more simple.

I use a combination of mouseover macros and Clique. I have my main healing spells on mouseover macros and my dispels and CDs on Clique macros.

Are you suggesting that using something like "shift+alt+right click" is not as easy or accurate, and limits movement? Because that is not the case at all.
I actually find that to be the case for myself. I heal using a combination of mouseover macros and click-casting using VuhDo. I found that trying to bind anything to ctrl or alt tended to confuse me and I ended up with my fingers on the wrong keys after a while (and running sideways into bad guys in the middle of the raid is a really good way to make people unhappy with you). Not to mention I would eventually forget what everything was bound to, which led to a lot of frustration. So while for you it may not limit movement and be just as accurate, you cannot state that as a fact. (c:

Apologies, YJMark. To me "because that is not the case at all" came across that you could not see how anyone else had problems with click-casting so many spells, and were presenting your opinion as fact.
I do very much agree that it's based on preference and one is not automatically easier than the other.

I find that for myself I attempted to use grid and clique, but never could remember the combos to use. Once I started using my G13 better and added mouseover macros, I found myself healing much better.
The most confusing issue for me is remembering where I have things arranged for the different classes, but have started to move some things to common keys, like my interrupt between the classes is on the same key, so I can just think ok interrupt and muscle memory takes over.
On any of these things though, I think the best advice I have here and elsewhere is try it and see which work better for you. One other bit of advice I have seen and believe is DO NOT attempt to do the change wholesale, incremental is much better of 2 to 4 key/click combo changes is the most any of us can handle at one time.

I'm not following what you mean. Are you suggesting that using something like "shift+alt+right click" is not as easy or accurate, and limits movement? Because that is not the case at all.
Yes. Can you do shift+alt+right click while strafing right and turning around? I am sure you say you can, but in practice it will be max 9 times out of 10.
