Currently I have a section for Single Target and AEO (although I do not have Inquisition yet)
I have Avenging Wrath at the beginning of the sections as well as Divine Plea for instant Holy Power, then my general rotation spells, then taunts - single and AEO, then Damage Reduction for myself and party, then heals...
Am I missing anything? Any suggestions, etc?
Here is my Current Setup:

Sucks I can't post a link to a screenshot of my setup -.-
Is there anyway I can show you guys how I have mine currently setup?
Yea I have never setup hotkeys before, so I will have to try that, I have always just clicked with a mouse lol
I guess my main hotkeys would be: Crusader Strike, Judgement, AW, Shield of Righteousness, Consecration, Holy Wraith and a few others...right?

Post a link, and I'll quote it for you.
You really should look into learning to use keybinds, it will make you a much better tank as you can react quicker, which directly translates into less time spent dead for you and your party members.

I also started out with mouse clicking at first but that was years ago now. I progressively used more and more key-bindings, while using both key-bindings and mouse interaction with spells.
I tried to keep it as simple as i could for myself, i started with manual ability activation without inquisition. it works quite ok.
To bind keys, a general tip is to make use of buttons and combinations comfortable for you
this is my layout which worked for me
Bar1(bottom):
, , , , , , , , , , ,
Bar2 (left)
Alternative I
:
, , , , , , , , ,
Bar2 (left)
Alternative II
:
, , , , , , , , , , ,

Alternative I
, 5 button mouse -
left , middle , right , back , forward
Since
is bound as secondary for Toggle Autorun i rebind
for secondary instead.
(primary key is
)
*
Alternative II
, Razer Naga -
left , middle , right , back , forward , to
due to bad placement on both
and
i skipped them for this mouse.
You can take advantage of CLCProt to get used to and train your rotations.

So true, learn how to keybind and you will never have to look at your bars again ;)
I've written a small (beginners)
keybinding guide
... might be useful but i'm still working on the site.

Joystiq did a great article on keybindings. I use a very similar keybind set and i click on only 4 abilities, the ones with 5m or more cooldowns.
http://wow.joystiq.com/2012/05/22/keys-to-pvp-keybinding-movement-and-clicking/
My bars look like this, 3 stacked up in the bottom middle of my screen:
................. <-- last 4 are not bound
............ <-- shift modifier on all the same keys as below
...................
