The most challenging class No.1: Warrior
Here comes the old saying: "A Warrior is as good as his gears" - I know this is not completely true, but it holds a lot of value, especially for a Warrior Tank (since the original post is focused on PvE).
Warrior is also the only class that requires rage when you are soloing/leveling (Druids will hardly go bear solo), and it is a completely different mechanism (from mana/energy) to manage, especially when you consider all 3 stances.
In PvP, Warrior can be really powerful when there is a healer (or Shaman for a WF totem) behind, but otherwise, his survivability is always in question.
No. 2: Shaman
Not only it's received the most nerfs since the launch of WoW, it is also the least mobile class (due to totems), with one of the most casts (again due to totems), and has the least CC/Anti-pushback skills.
Then again, like any other classes, Shaman can be powerful if played well, especially at mid-levels (20-50).
These are my 2cts, after spending decent amount of time in all classes in PvE and in PvP.
P/S - I'm surprised someone listed Mage as a challenging classes for PvE, because Mage is actually the easist for PvE (just consider how many mobs are either humanoids or beasts which are both sheepable....)
